The Big Brain is the leader of the Brain Spawn. He is a massive brain who lives in the Infosphere.
The Big Brain conducted several attacks on other planets by rendering their population stupid and then destroying the planet. The Big Brain helps Fry travel back to the past to stop himself from being frozen. The Brainspawn's goal was to gather all of the universe's information into the Infosphere. He was most likely destroyed when Fry blew up the Infosphere.
There may be several Big Brains, but only one was present at the invasion, and the Nibblonians implied that it was unique. It was instrumental in the attempted invasion and eradication of Earth and resided in New New York's Public Library during it, where it was fielded by Fry and Leela. The Big Brain has the ability to create fictional alternate realities (as found in books) and seal/trap its enemies in it. Fry chased the brain through several literary classics like Moby Dick, Tom Sawyer and Pride and Prejudice with the literary characters from the former two. While it was distracted, Fry made use of this ability and wrote a short story of its own, thus shaping a fictional reality himself in which the Big Brain became trapped and did whatever he wrote. It then left Earth, to which the Nibblonians seized the opportunity to destroy the invading Brain Spawn by devouring them. The Big Brain was later present in The Why of Fry when the Brain Spawn created the Infosphere, and successfully convinces Fry to briefly return to the year 1999. He was first killed when the Infosphere was destroyed, and, from the afterlife dimension, convinced Fry to return to 1999. However, it can be assumed that he did not leave the afterlife the second time Fry destroyed the Infosphere after returning from 1999.
Like all other Brain Spawn, the Big Brain can stupefy all animals with the exception of Fry as he lacks Delta Brainwave. It also has the ability to float.

- Conicidentally, Marurice LaMarche has also voiced a character named Brain in the TV series Pinky and the Brain.
- In The Why of Fry, it is revealed that is he is not largest of Brain Spawn. Despite being called "Big Brain," the most humongous brain is a brain who is known to some Futurama fans as "Huge Brain," though this Giant-like brain merely acts as a gigantic enforcer for the Big Brain.
